No study has tested whether NMN breaks a fast. For most people in the US who fast for weight or routine, the safest answer is simple: take NMN with water when you break the fast. That is what the Wellness Nest label advice for AgeAura says, and it removes the question.
Below are 6 rules for fasting days. Each one says what is known, what is not, and where the facts come from. For the general timing question, read our guide on when to take NMN. For trial doses, see how much NMN per day.
Know That No Trial Has Tested NMN During a Fast
We searched Europe PMC for randomized trials of NMN combined with intermittent fasting, time-restricted eating or a fasted state. The search returned 0 trials.
So any page that says NMN "does not break a fast" is giving an opinion. Nobody has measured it. Treat that as the starting point for every rule below.
Read the Label: A Capsule Is More Than NMN
Many "NMN and fasting" articles assume pure NMN powder. Most capsules hold more than that. Per 2-capsule daily serving, AgeAura Pure NMN + TMG NAD+ Complex lists:
| Ingredient | Amount per 2 capsules |
|---|---|
| NMN | 500 mg |
| TMG (betaine anhydrous) | 250 mg |
| Quercetin (Sophora japonica) | 150 mg |
| Trans-resveratrol (Polygonum cuspidatum) | 100 mg |
| Apigenin | 50 mg |
| Folate (L-5-MTHF) | 400 mcg DFE |
| Spermidine (wheat germ) | 1 mg |
| Capsule shell | Vegetable cellulose |
Source: the AgeAura product page. The panel lists no sugar and no calories. It does contain wheat, from the spermidine source. If your fast has strict rules, judge the whole capsule, not only the NMN.
Take It When You Break the Fast
The AgeAura product page gives direct advice for this case: take 2 capsules daily at the same time, and "if you fast, take with water when you break the fast."
This is the low-risk choice. Your fasting window stays clean, and the dose still lands at a fixed time each day. If you eat from noon to 8 p.m., that means noon.

What US Trials and Reviews Show About Timing and Blood Sugar
The human data on NMN and glucose come from people who were not fasting for a diet. Here is what the main studies tested:
- Irie 2020. 10 healthy men took single doses of 100, 250 or 500 mg. The doses were safe, and blood glucose dropped but stayed in the normal range.
- Yamaguchi 2024. 11 healthy middle-aged men took two 125 mg capsules once daily before breakfast for 8 weeks. It was well tolerated and NAD+ in blood cells rose. This was an open-label study with no placebo group.
- Yoshino 2021. In a 10-week randomized trial, NMN improved muscle insulin sensitivity in postmenopausal women with prediabetes who were overweight or obese.
- 2026 meta-analysis. Across 15 randomized trials (250 to 2,000 mg a day, 14 days to 24 weeks), NMN did not raise adverse events and did not change fasting glucose, HbA1c or body weight.
The Yamaguchi dose was taken before breakfast, after an overnight fast. That shows people have taken NMN on an empty stomach without problems in a trial. It does not show what NMN does to a longer fast.
Keep the Fasting Window to Calorie-Free Drinks
Harvard Health says that taking medications does not break a fast, and neither do calorie-free drinks like water or black coffee. It also advises people who fast to stay hydrated with calorie-free beverages.
A supplement is not a medication, so that statement does not settle the NMN question. It does give you a clear rule for the window itself: water, black coffee or plain tea. Save the capsules for your first meal.

Check With Your Doctor if You Fast on Medication
Harvard Health warns that fasting may be dangerous with certain medications, and that people with diabetes or heart disease should talk to their physician first. The AgeAura product page adds its own caution: if you are pregnant, nursing, under 18 or take medication, talk to a healthcare provider before you start.

OUR VERDICT
Nobody knows if NMN breaks a fast, because no trial has tested it. Do not guess. Keep the fasting window to water, black coffee or plain tea, and take your 2 capsules with water at your first meal, every day at the same time.

Does NMN break a fast?
No one knows for sure. No randomized trial has tested NMN during a fast. If you want a clean fast, take it with water when you break the fast, as the AgeAura label advice says.
Can I take NMN on an empty stomach?
In the Yamaguchi 2024 trial, men took NMN once daily before breakfast for 8 weeks, and it was well tolerated. That was a small study without a placebo group. If a capsule upsets your stomach, take it with food.
Does NMN spike insulin?
The trials we found do not show that. Single doses of up to 500 mg lowered blood glucose within the normal range, and a 2026 meta-analysis found no change in fasting glucose or HbA1c. None of these studies was done during a diet fast.
Can I take NMN during intermittent fasting?
Yes, if you time it to your eating window. Take 2 capsules with water at your first meal each day. Talk to your doctor first if you take medication or have diabetes or heart disease.
