NMN: 500 mg

NMN is the main ingredient. Its best-known human result is a 2021 randomized, placebo-controlled, double-blind trial in Science. In postmenopausal women with prediabetes who were overweight or obese, 10 weeks of NMN increased muscle insulin sensitivity and signaling.

What is not proven: that result came from one group of women with a metabolic problem. It does not show that NMN slows aging in healthy adults. Trial doses are covered in our NMN dose guide.

TMG (Betaine): 250 mg

TMG is betaine. It gives a methyl group to homocysteine and turns it back into methionine. A 2013 meta-analysis of 5 trials in healthy adults found that at least 4 g a day of betaine for 6 to 24 weeks lowered homocysteine by 1.23 μmol/L on average.

A 2002 trial in 42 obese adults gave 6 g a day for 12 weeks. Homocysteine went down, but body weight and body composition did not change. LDL cholesterol was higher in the betaine group.

What is not proven: those trials used 4 to 6 g. The AgeAura label has 250 mg, which is far below that range. Read it as a methyl-support ingredient, not a homocysteine treatment.

Folate as L-5-MTHF: 400 mcg DFE

Folate works next to TMG. The enzyme methionine synthase needs folate, in the form 5-methyltetrahydrofolate, plus vitamin B12 to convert homocysteine to methionine. When folate is low, homocysteine builds up.

The adult RDA is 400 μg DFE a day (600 in pregnancy, 500 while breastfeeding). The label dose matches the adult RDA. The upper limit of 1,000 μg a day applies to folic acid from supplements and fortified foods, so count any multivitamin you already take.

What is not proven: a benefit beyond normal folate status.

Trans-Resveratrol: 100 mg

Resveratrol is the most-studied extra. The Linus Pauling Institute reviewed the trials and found little or no metabolic benefit in overweight or obese people who did not already have a clear metabolic problem. Our NMN and resveratrol guide covers the trials in detail.

What is not proven: a clear benefit in healthy adults.

Quercetin: 150 mg

A 2016 meta-analysis pooled 7 trials with 587 patients. Quercetin lowered systolic blood pressure by 3.04 mmHg and diastolic by 2.63 mmHg. The effect showed up mainly at 500 mg a day or more. Below 500 mg a day, it was not significant.

What is not proven: any blood pressure effect at 150 mg.

Apigenin: 50 mg

Apigenin is on the label because of a 2013 lab study. Apigenin and quercetin inhibited CD38, an enzyme that uses up NAD+, and raised NAD+ in cell cultures and obese mice.

What is not proven: that this happens in people. We found no human trial of apigenin for NAD+.

Spermidine: 1 mg (From Wheat Germ)

The SmartAge trial gave 100 adults aged 60 to 90 with self-reported memory decline 0.9 mg a day of spermidine from wheat germ, or placebo, for 12 months. Memory did not change significantly (P = .47). Side effects were similar in both groups.

What is not proven: a memory or aging benefit.

What Is Not Proven About the Full Formula

We searched for a randomized human trial of NMN with betaine, resveratrol, quercetin, apigenin or spermidine. We found reviews, not trials. So nobody has shown that this combination works better than NMN alone. Also, three ingredients (TMG, quercetin, spermidine) sit below or near the doses where human trials looked.

AgeAura is sold by Wellness Nest at wellnessnest.co, where it sits in the best sellers collection. The product page states that it is made in a USA cGMP-certified partner facility, with non-GMO vegan capsules. It also says batch documentation will be published on the page when it is available, so third-party test results are not posted yet.
